In vivo imaging of injected differentiated cells and stem cells in a pig sphincter and establishment of an in vivo small animal model to determine cell mobility and retention time

Cross-sectional project 1 aims to develop and provide the consortium with a small-animal and clinical-translational imaging platform (positron emission tomography (PET), optical imaging (OI), magnetic resonance imaging (MRI) and PET/MRI). In addition, our expertise in iron-, fluorescent and radiolabelling will be brought in to establish methods that enable the visualisation of therapeutic mesenchymal stem cells (MSCs) and validate them in vitro and in vivo using rat and pig animal models. Non-invasive imaging will be used to quantitatively determine the temporal distribution (trafficking) and accumulation (homing) of the labelled MSCs. MRI will be used to examine the pigs in order to meet the translational aspect necessary for future application in human patients. In the field of regenerative medicine and cell-based regeneration, the combination of PET and MRI can deliver important elements related to improved detection of cells. The new PET/MRI method will therefore be used in the rat model and assessed for its suitability in preclinical research. PET/MRI is able to precisely correlate the enrichment of a radiolabelled biomarker with a specific morphological structure.
