Functional analysis of myocytes differentiated in vitro

While stem cell-differentiated cells are often quite homogeneous with regard to the expression of cell-type specific intracellular marker proteins, they differ considerably in their functional properties. However, these properties are crucial for the integration of the stem cell-differentiated cells into tissue according to their appropriate function.

Q2 characterises and compares the cell populations generated by myogenic differentiation from different stem cell sources in subproject TP1 with regard to their functional parameters. This is achieved through electrophysiological detection of so-called ‘marker ion flows’ typical for adult myocytes. The goal is to develop a reproducible, standardised differentiation method that results in an optimal functional homogeneity of the different myogenic cell populations.

In vitro co-culture approaches will subsequently be used to assess the relationship between the functional differentiation state of the stem cell-differentiated myocytes and their functional integration into existing muscle tissue. We expect these investigations to provide us with important information on which degree of maturation of the differentiated myocytes is suitable for therapeutic application.
